Hospedeiros alternativos de fitonematoides podem causar restrição nutricional e afetar a capacidade de infecção e fertilidade dos nematoides. Nesse trabalho, avaliou-se o efeito da planta hospedeira na viabilidade do inóculo de H. glycines raça 3 e a sobrevivência desse inóculo no solo. O nematoide foi multiplicado em feijoeiro resistente (linhagem L2300), suscetível (cv. Ouro) e soja suscetível (cv. FT-Cristalina), e as suspensões de ovos foram preparadas a partir de fêmeas coletadas nesses hospedeiros. Para avaliar a penetração dos juvenis de segundo estádio e o desenvolvimento do nematóide até a fase adulta, suspensões de 2000 ovos foram colocadas em plantas de soja 'FT-Cristalina' (planta teste). Aos 10 dias após a inoculação, avaliou-se o número de juvenis que penetraram nas raízes. O número de fêmeas por sistema radicular foi avaliado aos 30 dias. O inóculo obtido a partir da linhagem L2300 resultou no menor número de juvenis e de fêmeas no interior das raízes da planta teste. O inóculo obtido na soja foi o que resultou na maior penetração e desenvolvimento do nematoide. Para avaliar a sobrevivência do inóculo, cistos de H. glycines produzidos nos diferentes hospedeiros permaneceram no solo, sem planta, por 18 meses. O menor percentual de ovos que sobreviveram no solo foi originário de cistos multiplicados em feijoeiro, da linhagem L2300 ou cv. Ouro. Conclui-se que o inóculo de H. glycines produzido em feijoeiro, resistente ou suscetível, apresenta menor viabilidade e sobrevivência no solo.

Alternative hosts of phytonematodes can cause nutritional restriction and affect infectivity and fertility of the nematode. In this study, we evaluated the effect of the host plant on inoculum viability of Heterodera glycines race 3 and survival of this inoculum in soil. The nematode was multiplied in resistant (line L2300) and susceptible (cv. Ouro) beans and susceptible soybeans (cv. FT-Cristalina), and the egg suspensions were prepared from females collected on these hosts. To evaluate penetration of second stage juveniles and development of the nematode to the adult phase, suspensions of 2,000 eggs were placed in soybean 'FT- Cristalina' plants (test plant). At ten days after inoculation, we evaluated the number of juveniles that penetrated the roots. The number of females per root system was evaluated at 30 days. The inoculum obtained from line L2300 resulted in fewer juveniles and females inside the roots of the test plant. The inoculum obtained from soybean resulted in higher penetration and development of the nematode. To evaluate the survival of the inoculum, cysts of H. glycines produced in the different hosts remained in the soil, without plants, for 18 months. The lower percentage of eggs that survived in the soil was originated from cysts multiplied in beans, line L2300 or cv. Ouro. It can be concluded that the inoculum of H. glycines produced in bean plants, resistant or susceptible, has lower viability and survival in the soil.

A esporulação de Alternaria solani in vitro pode ser escassa e tende a reduzir após sucessivas repicagens. Conduziram-se vários experimentos para avaliar o efeito da umidade, injúria da colônia, qualidade de luz e fotoperíodo, bem como do meio de cultura na produção de conídios. Adaptou-se a técnica bifásica para induzir esporulação, por meio de injúria de micélio e desidratação do meio de cultura. Obtiveram-se melhores resultados quando colônias cresceram em meio V8 a 25ºC, no escuro, sob agitação durante sete dias; a massa de micélio foi triturada e a suspensão depositada em meio BDA (pH 6,5) em placas de Petri, incubando-se a 25 ± 2ºC, sob luz negra com 12 h de fotoperíodo. Validou-se o protocolo para 30 isolados de A. solani distintos quanto ao hospedeiro, local de origem, idade e forma de armazenamento. Quantificaram-se a produção, germinação e infectividade de conídios. Dos 30 isolados, 75% esporularam e a germinação mínima de conídios foi de 68%. Todos os isolados foram patogênicos em seus respectivos hospedeiros. Avaliou-se, também, o efeito de repicagens sucessivas na esporulação. Todos os isolados produziram conídios após seis repicagens sucessivas.

Sporulation of Alternaria solani can be scarce and is often reduced when the fungus is cultivated in vitro. A series of experiments were conducted to assess the effects of moisture, mycelial wounding, light quality and photoperiod, and culture media on conidial production. A procedure to induce sporulation based on mycelial wounding and dehydration was adapted and validated. Best results were obtained when fungal colonies were grown in V8 medium at 25ºC in the dark with agitation for seven days; the mycelium mass was ground, poured into potato dextrose agar (pH 6.5) in plates, and incubated at 25 ± 2ºC under near ultraviolet light and 12 h-photoperiod. The procedure was validated with 30 isolates of A. solani from different hosts, sampling locations, ages, and storage conditions. Conidial production, germination, and infectivity were quantified. Seventy-five percent of the isolates sporulated and the lowest germination rate was 68%. Inoculations of conidial suspension of all isolates resulted in lesions on tomato and potato plants. The effect of subculturing on A. solani sporulation was also assessed. After six subcultures, every seven days, there was no reduction in sporulation of colonies that were induced with the protocol.

A variabilidade de 53 isolados de Myrothecium roridum, obtidos de diferentes cultivos de meloeiro (Cucumis melo) do Estado do Rio Grande do Norte, foi estimada com base em variáveis relacionadas à: severidade do cancro-de-mirotécio nas cultivares AF-682 e Orange Flesh, fisiologia do patógeno (taxa de crescimento micelial - TCM - e esporulação - ESP) e sensibilidade ao fungicida fluazinam (ICM). Todos os isolados foram patogênicos às cultivares AF-682 e Orange Flesh, porém, nessa última houve maior discriminação entre isolados, quanto às variáveis mensuradas. A variabilidade entre os isolados foi alta quando se analisaram as variáveis medidas, com exceção de TCM e sensibilidade a fungicidas (todos sensíveis). Não foi verificada correlação significativa da SEV com as demais variáveis. A variação entre isolados dentro de município (60%) foi maior que a variação entre municípios. Provavelmente, há alta variabilidade genética nas populações de M. roridum e não foi possível detectar associações entre as variáveis analisadas e localidades amostradas.

The variability of 53 isolates of Myrothecium roridum obtained from melon (Cucumis melo) fields in the State of Rio Grande do Norte (Brazil) was estimated based on: severity of Myrothecium stem canker in the cultivars AF-682 and Orange Flesh, the pathogen's physiology (mycelium growth rate - MGR - and sporulation - SP) and its sensitivity to the fungicide fluazinam. All isolates were pathogenic to AF-682 and Orange Flesh; however, better discrimination of pathogen variability was achieved in the latter. Variation among isolates was high when assessing all variables, except for MGR and sensitivity to fungicides (all sensitive). There was no correlation between SEV and any other variables. The variation among isolates within municipalities (60%) was higher than the variation among municipalities. There is evidence of high genetic variability in the populations of M. roridum, but no association between the assessed variables and locations was detected.

A capacidade de dois isolados bacterianos obtidos do filoplano de tomateiro, em controlar a requeima (Phytophthora infestans) em condições de campo, foi comparada com o fungicida chlorothalonil (1,5 g l-1) e água (controle). Foi observada uma redução de 55% na intensidade da doença em folhas da do terço mediano da planta e de 62% nas folhas superiores, quando pulverizadas com o antagonista UFV-STB 6 (Novosphingobium capsulatum) em relação ao tratamento controle. O isolado UFV-IEA 6 (Bacillus cereus) foi capaz de reduzir a intensidade da doença em 55%, somente nas folhas do terço superior das plantas. As plantas pulverizadas com o isolado UFV-STB 6 também apresentaram redução significativa no percentual de frutos sintomáticos. Os resultados demonstram que os dois isolados bacterianos têm potencial no controle da doença.

The capacity of two bacteria isolated from the tomato phylloplane to control late blight (Phytophthora infestans) was investigated in the field, and compared against the effectiveness of spraying with the fungicide chlorothalonil (1.5 g a.i. L-1) or water (control). A 55% reduction in late blight intensity was observed in the leaves of the middle of the plant and 62% in those of the upper leaves when using the antagonist UFV-STB 6 (Novosphingobium capsulatum) as compared to the control. Isolate UFV-IEA 6 (Bacillus cereus) was able to reduce disease intensity by 55%, but only in the upper leaves of the tomato plants. Treatment with isolate UFV-STB 6 also led to a significant reduction in the percentage of fruits with late blight symptoms. The results demonstrate the potential of these two bacteria in controlling this disease.

Transformantes de Magnaporthe grisea resistentes a higromicina, obtidos principalmente por procedimentos REMI (Restriction Enzyme-Mediated Integration), foram avaliados quanto à habilidade em causar doença em arroz. A partir de testes de patogenicidade envolvendo 125 transformantes foi possível selecionar cinco mutantes com alterações consistentes na patogênese. Dois desses mutantes, T108 e T93, causaram poucas lesões em folhas de arroz, enquanto o mutante T251 não foi patogênico. A alteração na patogenicidade de T108 foi acompanhada pela menor capacidade de desenvolvimento in vitro. Quando inoculado em plantas de arroz, o mutante T41 apresentou agressividade reduzida, caracterizada por lesões arredondadas de tamanho limitado. Por sua vez, o período de incubação para o mutante T72 foi maior que o do isolado selvagem. Além disso, atraso considerável na germinação de conídios e formação de apressórios foi detectado em T72. Todos os mutantes cresceram em meio mínimo, indicando que as mutações não foram associadas à auxotrofia. Os mutantes T93 e T251 apresentam fenótipos semelhantes quando em cultura, caracterizados pela pigmentação marrom.

Hygromycin-resistant transformants of Magnaporthe grisea, generated mainly by REMI (Restriction Enzyme-Mediated Integration), were screened for their ability to infect rice plants. Out of 125 transformants five showed changes in pathogenicity. The T108 and T93 mutants caused few lesions in rice leaves, while the T251 mutant was non-pathogenic. The alteration in pathogenicity of T108 was accompanied by reduced development in culture. The T41 mutant caused small and limited round lesions. The incubation period of the T72 mutant was longer than that of the wild type. Furthermore, late germination and appresorium formation was detected in the T72 mutant. All mutants grew in minimum medium, suggesting that there were no auxotrophic-associated mutations. The T93 and T251 mutants presented similar phenotypes in culture, characterized by a brown-pigmented colony.

Em um processo de seleção de bactérias do filoplano de tomateiro (Lycopersicon esculentum) com potencial para o controle de doenças da parte aérea da cultura, diferentes métodos de isolamento foram utilizados visando obter isolados da população total, da população da superfície foliar e isolados que habitam locais protegidos do filoplano e/ou que toleram fatores de estresse. Foi testada a capacidade de 300 isolados em controlar in vivo, as doenças causadas por Alternaria solani, Pseudomonas syringae pv. tomato e Phytophthora infestans. Os testes foram repetidos para cada um dos antagonistas selecionados, estudando-se, também, a capacidade de controlar a mancha-bacteriana, causada por Xanthomonas vesicatoria. Os resultados demonstraram haver predomínio de antagonistas provenientes de folíolos do terço superior da planta de população total ou da superfície. Entretanto, o único antagonista selecionado, isolado de folíolos do terço inferior, foi obtido de locais protegidos do filoplano e/ou capaz de tolerar fatores de estresse.

In a process of selecting tomato (Lycopersicon esculentum) phylloplane bacteria with potential for controlling aerial diseases of this crop, different isolation methods were used to obtain the isolates from total leaf surface and also from protected sites of the phylloplane. Three hundred isolates were tested in vivo for their ability to control diseases caused by Alternaria solani, Pseudomonas syringae pv. tomato and Phytophthora infestans. The results demonstrated a predominance of antagonists in the total population originating on the surface of the leaflets harvested from the upper leaves of the plants. Only one selected antagonistic isolate obtained from the lower leaves was recovered from the protected sites of the phylloplane and/or was stress tolerant.
